Genome‐wide scan of long noncoding RNA single nucleotide polymorphisms and pancreatic cancer susceptibility

Genome-wide scan of long noncoding RNA (lncRNA) variants in 19,862 individuals identified five novel lncSNP associations with pancreatic ductal adenocarcinoma (PDAC) risk. The strongest association was rs7046076 (C allele, OR=1.13, 95% CI=1.09-1.18, P=9.73×10⁻⁹) in the NONHSAG053086.2/lnc-SMC2-1 lncRNA, which disrupts binding to hsa-mir-1256, regulating genes including CDKN2B and DAAM1 involved in cell cycle control and cell migration.

This functional study investigated the molecular mechanism of the osteoporosis GWAS variant USF3 c.3781C>A (rs1026364), which is associated with femoral neck BMD (P=4.1×10⁻¹⁰). The authors demonstrated that the c.3781A allele creates a functional binding site for hsa-miR-345-5p, which downregulates USF3 expression and inhibits osteogenic differentiation through both USF3-dependent and independent pathways, including direct targeting of RUNX3 and SMAD1.

CDK6 3'UTR polymorphisms alter the susceptibility to cervical cancer among Uyghur females

A case-control study of 306 cervical cancer cases and 310 healthy Uyghur female controls found that CDK6 3'UTR variants rs8179 and rs42033 reduced cervical cancer susceptibility (OR=0.60, 95% CI 0.37-0.99, p=0.043). rs8179, rs42032, and rs42033 showed stronger protective effects against high-grade cervical cancer (OR=0.35-0.43, p<0.01). CDK6 expression was significantly reduced in cervical tumors.
