A genetic stochastic process model for genome‐wide joint analysis of biomarker dynamics and disease susceptibility with longitudinal data
MethodsN=8,284Liang He et al.(2017)· Genetic Epidemiology

This paper proposes a novel genetic stochastic process model (GSPM) for genome-wide association studies of longitudinal biomarker data and disease susceptibility. Applied to the ARIC study (8,284 Caucasians, 22,101 BMI observations), the authors identified SNP-BMI interaction effects on type 2 diabetes risk, including rs11757677 in CDKAL1 (p=5.77e-07) showing significant interaction effects, and rs1551133 on 2q14.2 (p=6.70e-07) that reversed the BMI effect on T2D risk. The method jointly models biomarker dynamics and disease hazards, providing improved biological interpretation of genetic effects.

About CDKAL1

The protein encoded by this gene is a member of the methylthiotransferase family. The function of this gene is not known. Genome-wide association studies have linked single nucleotide polymorphisms in an intron of this gene with susceptibilty to type 2 diabetes. [provided by RefSeq, May 2010]
