This randomized controlled trial (N=3,819) assessed melanocortin 4 receptor (MC4R) locus variants and weight-loss response in the Diabetes Prevention Program. Twenty SNPs were genotyped across MC4R; the minor allele at rs1943218 was nominally associated with weight change at 6 months and 2 years, and eight SNPs modified treatment response to lifestyle or metformin interventions on weight loss or diabetes incidence.
