This study conducted an exhaustive genome-wide SNP-SNP interaction analysis in the ADNI dataset using the EPISIS machine learning algorithm to identify epistatic effects on brain volume. The authors identified a significant SNP pair interaction between rs1345203 and rs1213205 that was associated with ~5% greater temporal lobe volume in both ADNI (n=737) and independent QTIM (n=753) samples, with protective effects observed bilaterally in temporal and occipital brain regions.
