Genetic relationships between suicide attempts, suicidal ideation and major psychiatric disorders: A genome‐wide association and polygenic scoring study

This genome-wide association and polygenic scoring study examined genetic relationships between suicide attempts, suicidal ideation, and major psychiatric disorders across four cohorts (RADIANT n=2,023; GSK-Munich n=807; BACCs n=440; GENDEP n=747). No SNPs reached genome-wide significance in GWAS meta-analyses. Polygenic scores for major depressive disorder significantly predicted suicidal ideation (P=0.015, explaining 1% of variance) and suicide attempts in combined validation dataset (R²=0.3%, P=0.013), suggesting pleiotropy between psychiatric disorders and suicidality. Suicide attempt and ideation appear to have partially independent genetic architectures.
