Pathway‐analysis of published genome‐wide association studies of lung cancer: A potential role for the CYP4F3 locus

Pathway-based meta-analysis of 6 GWAS studies from the TRICL consortium (12,160 lung cancer cases and 16,838 controls) combined with Harvard GWAS data identified CYP4F3 rs4646904 (G>A) as a potentially functional SNP associated with lung cancer risk (OR=1.07, 95%CI=1.03-1.12, P=3.52×10-3). The variant showed stronger association in smokers (OR=1.13, P=6.70×10-5) and is predicted to influence splicing efficiency of CYP4F3, a gene involved in fatty acid metabolism.
