The dal-GenE trial tested whether dalcetrapib, a CETP inhibitor, reduces cardiovascular events in 6147 patients with acute coronary syndrome and the AA genotype at rs1967309 in the ADCY9 gene. The primary composite endpoint (cardiovascular death, resuscitated cardiac arrest, non-fatal myocardial infarction, or stroke) occurred in 9.5% of dalcetrapib-treated patients vs 10.6% of placebo-treated patients (hazard ratio 0.88, 95% CI 0.75-1.03, P = 0.12), which was not statistically significant, though a reduction in non-fatal myocardial infarction was observed (HR 0.79, 95% CI 0.65-0.96).

Adenylate cyclase is a membrane bound enzyme that catalyses the formation of cyclic AMP from ATP. It is regulated by a family of G protein-coupled receptors, protein kinases, and calcium. The type 9 adenylyl cyclase is a widely distributed adenylyl cyclase, and it is stimulated by beta-adrenergic receptor activation but is insensitive to forskolin, calcium, and somatostatin.
