This replication study confirms that TUB SNP rs2272382 is associated with increased obesity measures in postmenopausal women from a large UK twin cohort (N=2,694). In postmenopausal women homozygous for the minor allele, rs2272382 showed associations with BMI (beta=0.92 kg/m², p=0.036), waist circumference (beta=2.73 cm, p=0.013), and percent central fat (beta=2.43%, p=0.027), with significant interaction with menopausal status (p=0.036). The association was confirmed by sibling transmission disequilibrium test in discordant dizygotic twins.

About TUB

This gene encodes a member of the Tubby family of bipartite transcription factors. The encoded protein may play a role in obesity and sensorineural degradation. The crystal structure has been determined for a similar protein in mouse, and it functions as a membrane-bound transcription regulator that translocates to the nucleus in response to phosphoinositide hydrolysis.
