A genome-wide association study of 484 trauma-exposed individuals identified ten SNPs with suggestive associations (p < 10^-5) with dissociative symptoms (derealization and depersonalization) in PTSD. The strongest association was rs263232 (β = 1.4, p = 6.12 × 10^-7) in ADCY8, and rs71534169 (β = 1.63, p = 3.79 × 10^-6) in DPP6. No SNPs met genome-wide significance. These genes are implicated in synaptic plasticity, memory consolidation, and neuronal excitability, suggesting biological pathways underlying dissociative phenomena.
About ADCY8
Adenylate cyclase is a membrane bound enzyme that catalyses the formation of cyclic AMP from ATP. The enzymatic activity is under the control of several hormones, and different polypeptides participate in the transduction of the signal from the receptor to the catalytic moiety. Stimulatory or inhibitory receptors (Rs and Ri) interact with G proteins (Gs and Gi) that exhibit GTPase activity and they modulate the activity of the catalytic subunit of the adenylyl cyclase [provided by RefSeq, Jul 2008]
