This association study examined SNX24 genetic variants in 262 Kawasaki disease (KD) patients (76 with coronary artery aneurysm (CAA) and 186 without) compared to 1107 healthy controls. The SNX24 SNP rs28891 showed significant association with CAA formation (p=0.006), with CC and CT genotypes showing protective effects (OR=0.29, 95% CI=0.12-0.70 for CC genotype; OR=0.48, 95% CI=0.27-0.87 for CT genotype). Functional studies in endothelial cells demonstrated that SNX24 knockdown decreased proinflammatory cytokines IL-1β, IL-6, and IL-8.
