Each copy of the T allele at rs35744813, located 1,266 bp upstream of TAS1R3, reduces sucrose taste sensitivity by roughly 25%, with T/T homozygotes showing approximately 50% decreased sensitivity compared to C/C individuals. TAS1R3 encodes a subunit of the TAS1R2/TAS1R3 heterodimeric sweet taste receptor, and this promoter variant alters a cis-regulatory silencer element that lowers gene transcription. In a study of 144 individuals, rs35744813 alone explained ~16% of population variability in sucrose perception (Bonferroni-corrected p = 2 × 10⁻⁵).
