This association study examined inherited genetic variation in six immune/inflammatory response genes (ABL2, SEMA4D, ITGAL, C1QA, TIMP1, CDKN1A) previously identified in a blood RNA prognostic model for castration-resistant prostate cancer (CRPC). In a test cohort of 600 CRPC patients, four SNPs were significantly associated with overall survival: rs1318056 and rs1490311 in ABL2 (aHR 1.33, p=0.037 and aHR 1.27, p=0.030, respectively) and rs2073917 and rs3764322 in ITGAL (aHR 1.39, p=0.043 and aHR 1.30, p=0.010). The two ABL2 SNPs were associated with reduced ABL2 mRNA expression in blood (eQTL p=0.035 and p=0.015), validating their functional relevance.
About ITGAL
ITGAL encodes the integrin alpha L chain. Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain. This I-domain containing alpha integrin combines with the beta 2 chain (ITGB2) to form the integrin lymphocyte function-associated antigen-1 (LFA-1), which is expressed on all leukocytes. LFA-1 plays a central role in leukocyte intercellular adhesion through interactions with its ligands, ICAMs 1-3 (intercellular adhesion molecules 1 through 3), and also functions in lymphocyte costimulatory signaling. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
