Role of GABRA2 in Trajectories of Externalizing Behavior Across Development and Evidence of Moderation by Parental Monitoring

A longitudinal association study of 378 participants from the Child Development Project (white subsample) examined the relationship between GABRA2 variants and trajectories of externalizing behavior from ages 12-22. Two trajectory classes emerged: stable high externalizing and moderate decreasing behavior. Multiple GABRA2 SNPs showed significant association with trajectory class membership (ORs 2.1-2.7, p<.001 to p=.001). Importantly, parental monitoring significantly moderated the genetic effect, with high parental monitoring diminishing the association of GABRA2 with persistent externalizing behavior (interaction p-values .007 to .03).

About GABRA2

GABA is the major inhibitory neurotransmitter in the mammalian brain where it acts at GABA-A receptors, which are ligand-gated chloride channels. Chloride conductance of these channels can be modulated by agents such as benzodiazepines that bind to the GABA-A receptor. At least 16 distinct subunits of GABA-A receptors have been identified.
