The authors identified rs11352835 (A/- insertion/deletion in OAS1 exon 7) as a novel multiple sclerosis susceptibility variant through population genetics analysis in 660 MS cases and 503 controls (OR 1.27, p = 0.010, allelic model). While the previously reported splice-site variant rs10774671 shows neutral evolution, the downstream exon 7 region exhibits signatures of balancing selection, and tight linkage between the two SNPs may explain conflicting results across populations.

This case-control study of 304 samples (164 cancer, 140 control) found that the OAS1 rs2660 AA genotype was significantly associated with increased prostate cancer risk (OR=2.4, p=0.0006), while the GG genotype was protective (OR=0.066, p<0.0001), reducing risk by 92%. The association was consistent across both Caucasian and African American populations, with the AA genotype showing additive effects with age.
About OAS1
This interferon-induced gene encodes a protein that synthesizes 2',5'-oligoadenylates (2-5As). This protein plays a key role in innate cellular antiviral response, and has been implicated in other cellular processes like cell growth and apoptosis. Alternative splicing results in multiple transcript variants with different enzymatic activities. Polymorphisms in this gene have been associated with susceptibility to viral infection, including SARS-CoV-2, and diabetes mellitus, type 1.
