A two-stage case-control study (7,763 subjects) identified rs12740674 (C>T) in the miR-1262 enhancer as a novel lung cancer susceptibility locus in the Han Chinese population, with the risk T allele associated with increased lung cancer risk (OR=1.24, 95% CI 1.13-1.36, p=6.204×10⁻⁶ in meta-analysis). Functional studies showed the risk allele reduces miR-1262 expression through chromosomal looping, with miR-1262 acting as a tumor suppressor by targeting autophagy and oncogenic pathways (ULK1 and RAB3D).
