Genetic variation in the endocannabinoid system and response to Cognitive Behavior Therapy for child anxiety disorders

This study investigated genetic variation in the endocannabinoid system (CNR1, CNR2, FAAH genes) as a predictor of response to cognitive behavior therapy for childhood anxiety disorders in 1,309 children aged 5-17 years. Six SNPs showed nominal associations (P<0.05) with treatment response during follow-up: rs806365 (CNR1, P=0.004), rs2501431 (CNR2), rs2070956 (CNR2), rs7769940 (CNR1), rs2209172 (FAAH) associated with poorer response, and rs6928813 (CNR1) with better response. Only rs806365 survived multiple testing corrections in the fear-based anxiety disorder subset (P=0.0011). The authors conclude there is very limited evidence for genetic variation in endocannabinoid system genes predicting CBT treatment response.

Evaluation of common variants in CNR2 gene for bone mineral density and osteoporosis susceptibility in postmenopausal women of Han Chinese

This case-control study evaluated 39 SNPs in the CNR2 gene for association with bone mineral density (BMD) and osteoporosis in 1,032 postmenopausal osteoporosis patients and 2,089 healthy Han Chinese controls. Two SNPs (rs4237 and rs2501431) were significantly associated with osteoporosis, with rs4237 showing p=0.000515 and rs2501431 showing p=0.000441, both with odds ratios around 1.2. Haplotype analysis identified five haplotypes with significant associations, particularly HAP9 which showed 4-fold increased frequency in cases (p<0.001).

About CNR2

The cannabinoid delta-9-tetrahydrocannabinol is the principal psychoactive ingredient of marijuana. The proteins encoded by this gene and the cannabinoid receptor 1 (brain) (CNR1) gene have the characteristics of a guanine nucleotide-binding protein (G-protein)-coupled receptor for cannabinoids. They inhibit adenylate cyclase activity in a dose-dependent, stereoselective, and pertussis toxin-sensitive manner. These proteins have been found to be involved in the cannabinoid-induced CNS effects (including alterations in mood and cognition) experienced by users of marijuana.
