Serum BLyS levels increase after rituximab as initial therapy in patients with follicular grade 1 non‐Hodgkin lymphoma

This study measured serum B-lymphocyte stimulator (BLyS) levels before and after rituximab treatment in 30 patients with previously untreated follicular Grade 1 non-Hodgkin lymphoma. Serum BLyS levels increased significantly after rituximab therapy (mean 4.47 ng/ml pre-treatment to 10.75 ng/ml post-treatment, P=0.0001), independent of four polymorphisms in the BLyS gene (TNFSF13B: rs12428930, rs12583006, rs1224141, rs9514828). While the increase in BLyS did not correlate with time to progression or overall survival, patients with higher post-rituximab BLyS levels were more likely to have complete response.

About TNFSF13B

The protein encoded by this gene is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. This cytokine is a ligand for receptors TNFRSF13B/TACI, TNFRSF17/BCMA, and TNFRSF13C/BAFFR. This cytokine is expressed in B cell lineage cells, and acts as a potent B cell activator. It has been also shown to play an important role in the proliferation and differentiation of B cells. Alternatively spliced transcript variants encoding distinct isoforms have been identified.
